What Water to Mix With Peptides

Choosing the right solvent is critical for peptide reconstitution. This guide covers the most common options and when to use each one.

Reconstitution Solvents

The solvent you choose for peptide reconstitution directly impacts stability, solubility, and experimental results. Here are the most common options used in research settings.

Solvent
Best For
Key Notes
Bacteriostatic Water (BW)
Most general-purpose reconstitution
Contains 0.9% benzyl alcohol as preservative. Allows multi-use vial access. Most common choice for lyophilized peptides. Sterile and widely available.
Sterile Water (SW)
Single-use applications
No preservative -- must be used immediately or discarded. Preferred when benzyl alcohol may interfere with the assay. Not suitable for multi-dose vials.
Normal Saline (0.9% NaCl)
Physiological studies, nasal sprays
Isotonic solution that closely matches physiological conditions. Used in Stemcode nasal spray formulations. Helps maintain peptide stability through proper osmolarity.
Acetic Acid (0.1%)
Hydrophobic or basic peptides
Weak acid that improves solubility for peptides with high isoelectric points. Useful when peptides won't dissolve in neutral water. Dilute further after initial dissolution.
DMSO
Very hydrophobic peptides
Last resort for peptides insoluble in aqueous solvents. Dissolve in a minimal amount of DMSO first, then dilute with aqueous buffer. Final DMSO concentration should stay below 10%.

Reconstitution Technique

Follow these steps for proper peptide reconstitution in a research setting.

01

Allow peptide to reach room temperature

Remove the vial from cold storage and let it equilibrate to room temperature for 15-20 minutes. This prevents condensation from entering the vial when opened and ensures even dissolution.

02

Add solvent slowly along the vial wall

Using a sterile syringe, add your chosen solvent slowly against the inside wall of the vial -- not directly onto the lyophilized cake. This prevents foaming and protein aggregation from mechanical stress.

03

Swirl gently -- never vortex

Allow the peptide to dissolve by gently swirling or rolling the vial between your palms. Never vortex or shake aggressively. Most peptides will dissolve within 1-5 minutes. If not fully dissolved, let it sit and swirl again.

04

Verify the solution is clear

A properly reconstituted peptide solution should be clear and colorless. If the solution remains cloudy or has visible particulates, the peptide may be damaged or the wrong solvent was used.

05

Aliquot and store

Divide the reconstituted solution into single-use aliquots. Store at 2-8°C for short-term use (2-4 weeks) or -20°C for longer storage. Label each aliquot with the peptide name, concentration, date, and lot number.

Pre-Formulated Nasal Sprays

Stemcode Peptides nasal sprays are pre-formulated and ready to use -- no reconstitution required. Each spray is prepared with:

  • Normal Saline (pH 6.0) as the base solution
  • Benzalkonium Chloride (BKC) as a preservative to inhibit microbial growth
  • Lab-grade distilled water for consistent purity

This eliminates the variability introduced by manual reconstitution and provides optimized stability from the manufacturer.
