Semax / DOSAGE RESEARCH GUIDE

Semax Dosage: How Much Is Used in Research

Published Semax studies span 200-600mcg daily intranasal in cognitive work to 12-18mg in stroke trials. What the literature used, and what it leaves open.

THE 30-SECOND OVERVIEW

Semax dosage research quick start

Semax has one of the more unusual dosing literatures among research peptides: nearly all of it is intranasal, most of it is Russian, and the doses span two orders of magnitude depending on what was being studied. This article summarises the dosing used in published Semax research. It is a description of what investigators did, not a protocol recommendation.

What Semax Is

Semax is a synthetic heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro) built from the ACTH(4-10) fragment of adrenocorticotropic hormone with a Pro-Gly-Pro tail added at the C-terminus. The PGP addition is the design decision that made the molecule practical: the bare ACTH(4-10) fragment is degraded within minutes, while the stabilised analogue persists long enough to act.

Two mechanistic points matter for reading its dosing literature. First, although Semax derives from ACTH, it retains no meaningful corticotropic activity: studies do not report cortisol stimulation, which separates its effects from a stress-axis response. Second, the best-characterised downstream effect is upregulation of BDNF and its receptor trkB in the hippocampus, demonstrated in animal work, which is the usual mechanistic frame for the cognitive findings.

Semax was developed at the Institute of Molecular Genetics in Moscow and has held Russian regulatory approval since the 1990s as a nasal solution in two strengths (0.1% and 1%), and that two-strength split is the key to understanding the dose ranges below.

The Dosing Used in Published Research

The literature divides cleanly into two regimes that track the two solution strengths.

The low-dose cognitive and asthenia studies used the 0.1% solution. Daily totals in this line of work generally fall in the 200-600mcg range, administered intranasally in divided doses across the day. The early human work that established the nootropic-like signal, attention and memory tasks in healthy operators under fatigue, used single intranasal administrations in the low hundreds of micrograms and measured effects over hours. Courses in this regime are short: days to a few weeks, not continuous administration.

The high-dose neurological studies used the 1% solution. The acute ischaemic stroke trials led by Gusev and Skvortsova administered 12mg or 18mg per day intranasally, in divided doses, for roughly the first ten days after stroke onset. These totals are twenty to sixty times the cognitive-study doses. Same molecule, same route, an entirely different research question.

The spread is the single most important fact in the Semax dosing literature. A figure quoted without its regime is close to meaningless: 300mcg and 18mg are both "doses used in research," from studies asking different questions in different populations.

Why Intranasal

Semax is administered intranasally in essentially all of the published work, and the reason is pharmacokinetic rather than convenience. Peptides of this class have negligible oral bioavailability, and the nasal route offers direct access toward the CNS compartment that the research questions target. Animal distribution work supports rapid uptake after nasal administration.

This is also why the route is not interchangeable in the literature: the published doses are intranasal doses. There is no meaningful oral Semax dosing literature to compare them against, and injectable administration barely features in the published human work.

What the Literature Does and Does Not Establish

The honest reading of the Semax evidence base has clear limits, and they are worth stating as plainly as the doses.

Most of the clinical literature is Russian-language, from a period when trial registration and reporting standards differed from current Western norms. The stroke trials enrolled hundreds of participants, but independent replication outside Russia is thin. The cognitive studies are small: dozens of participants, short durations, surrogate endpoints like attention-task performance and EEG measures.

Course lengths in the literature are short. The stroke protocols ran about ten days; cognitive protocols ran days to weeks. There is no published long-term continuous administration data, which means duration questions are simply unanswered rather than answered reassuringly.

The two regimes were also studied in different populations (acute stroke patients under hospital supervision versus healthy adults in task studies), and results from one population do not transfer to the other by assumption.

Reconstitution and Handling

Semax in nasal spray format is supplied ready to store under refrigeration, and the vial format is supplied lyophilised for reconstitution. The handling consideration that matters most for research use is concentration bookkeeping: because the literature's two regimes differ by more than an order of magnitude, a solution-strength error reproduces the difference between a cognitive-study dose and a stroke-trial dose. Recording the actual concentration in use is the discipline the two-regime literature forces.

For researchers comparing the molecule against its frequent counterpart, our Semax vs Selank comparison covers how the two peptides' evidence bases differ, and the Semax research overview collects the study register. Lots for both formats are covered in our lab results; researchers sourcing material can buy Semax nasal spray in the 10mg format used for spray-based work.

The Takeaway

Published Semax dosing is intranasal and regime-dependent: 200-600mcg daily from the 0.1%-solution cognitive literature, 12-18mg daily from the 1%-solution stroke trials, both in divided doses over short courses. No published figure floats free of its regime, and the literature's main gaps, long-term administration and non-Russian replication, are gaps rather than answers.
