Also Known As: Ibutamoren, MK-0677

MK-677 is also known as Ibutamoren or MK-0677 — these names refer to the same orally active small-molecule ghrelin receptor (GHS-R1a) agonist studied in growth hormone and IGF-1 research. It is a small molecule, not a peptide and not a SARM. For research use only.

Description

MK-677, also known as Ibutamoren or MK-0677, is an orally active small-molecule growth hormone secretagogue built on a spiroindoline scaffold. It acts as a ghrelin mimetic — an agonist of the growth hormone secretagogue receptor GHS-R1a. In published clinical research, once-daily oral administration enhanced pulsatile growth hormone secretion and raised serum IGF-1 concentrations in a dose-dependent manner. Unlike most compounds in this catalog, MK-677 is a small molecule, not a peptide. Supplied as 15mg tablets, 60 per container. For research use only.

MK-677 is also stocked as a measured oral solution, MK-677 Liquid at 25mg per mL, for dose-response designs that draw a volume rather than split a tablet.

Research Background

MK-677 has one of the deepest clinical literatures of any growth hormone secretagogue. Chapman et al. (1996) reported that daily oral MK-677 enhanced pulsatile GH release and restored IGF-1 to young-adult ranges in healthy older subjects. A 2-year randomized trial in healthy older adults (Nass et al., Annals of Internal Medicine, 2008) found increased GH and IGF-1 levels and increased fat-free mass, alongside increased fasting glucose and decreased insulin sensitivity. Other trials examined nitrogen balance during caloric restriction, bone-turnover markers and bone mineral density, sleep architecture, and appetite-related ghrelin signaling.

This product is intended strictly for laboratory research use only and is not for human consumption, clinical use, diagnostic use, or any therapeutic application.

YearStudyDesignWhat it found
1996 Chapman et al., J Clin Endocrinol Metab 32 healthy adults aged 64-81; placebo, 2, 10 or 25mg once daily; 14- and 28-day periods 25mg raised mean 24-hour GH 97% and IGF-1 from 141 to 265 µg/L by 4 weeks; fasting glucose rose 5.4 to 6.8 mmol/L; cortisol unchanged; prolactin +23% within normal range.
1996 Copinschi et al., J Clin Endocrinol Metab 9 healthy young men; placebo, 5 or 25mg at bedtime; 7-day crossover GH pulse frequency rose through added low-amplitude pulses; IGF-1 rose dose-dependently; 24-hour cortisol unchanged apart from timing.
1997 Copinschi et al., Neuroendocrinology 8 young and 6 older adults; 2, 5 or 25mg at bedtime; 7- and 14-day periods In the young, stage IV sleep +50% and REM +20%; in adults 65-71, REM +50% and shorter REM latency.
1998 Murphy et al., J Clin Endocrinol Metab 8 volunteers on 18 kcal/kg/day; 25mg once daily for the last 7 of 14 days; crossover Nitrogen balance +0.31 g/day on MK-677 against -1.48 g/day on placebo; IGF-1 restored.
1998 Svensson et al., J Clin Endocrinol Metab 24 obese men; 25mg daily or placebo; 8 weeks IGF-1 +40%; fat-free mass up; total and visceral fat unchanged; oral glucose tolerance impaired at 2 and 8 weeks.
1999 Murphy et al., J Bone Miner Res 187 adults 65+ across three placebo-controlled studies of 2-9 weeks; 5-50mg Bone resorption marker NTx +10-17% at 2 weeks; osteocalcin +29% at 9 weeks; IGF-1 +55-94%.
2001 Codner et al., Clin Pharmacol Ther 18 prepubertal children with GH deficiency; 0.2 or 0.8mg/kg/day; 7-8 days 0.8mg/kg raised peak GH, IGF-1 and IGFBP-3; no change in prolactin, glucose, thyroid hormones, cortisol or insulin.
