What is SS-31 (Elamipretide)?

SS-31, also known as Elamipretide or MTP-131, is a synthetic peptide engineered for research into mitochondrial structure and cellular bioenergetics.

It’s best known for its ability to selectively localize to the inner mitochondrial membrane, where it interacts with cardiolipin — a phospholipid involved in maintaining mitochondrial architecture.

When administered via nasal spray, the peptide is positioned for research investigating nasal-mucosal absorption pathways, which are commonly studied for compounds of neurological interest.

Mitochondrial Structure & Cristae Stability

SS-31 has an affinity for cardiolipin, a phospholipid found on the inner mitochondrial membrane. In cell and animal studies, this interaction has been shown to help preserve cristae structure (the inner folds of the mitochondria), especially under conditions of oxidative or metabolic stress. Having more inner folds in the mitochondria means there’s more surface area for energy-producing enzymes to work. 

These studies indicate SS-31 may stabilize mitochondrial morphology during experimental stress exposure [1].

ATP Generation & Bioenergetic Support

Some studies suggest that when SS-31 helps maintain mitochondria during stress, the cell can produce energy more efficiently. Because of this, researchers are studying SS-31 in experiments on low-energy conditions, cell fatigue, and stress. For instance, in one study, the researchers found that this mitochondrial-targeted peptide rapidly improved mitochondrial energy output and skeletal muscle performance in aged mice [2,3].

Neuroprotection Research

Some early research shows that SS-31 may help support how brain cells handle stress. Because of this, scientists are studying it in mouse and rat models of aging brain cells and conditions in which cells are easily damaged [4, 5].
